摘要:
地衣芽胞杆菌发酵产杆菌肽的过程中,菌体前期生长和代谢的强度会影响杆菌肽的合成。为了提高效价,在20 L发酵罐水平研究变温控制策略对效价合成的影响,发现:37℃发酵温度(对照)下的效价在21 h到达最高点(962 UmL),平均效价合成速率为45.33 U(mL·h)。通过降低发酵培养温度(35.5℃、34℃、32.5℃和31℃),可以降低菌体生长速率和糖耗速率,虽然会降低平均效价合成速率,但延长了效价合成时间。在34℃发酵温度下以37.66 U(mL·h)的平均效价合成速率至29 h到达效价最高点(1092 UmL),比37℃条件下提高了13.5%。基于降温控制策略进行18 h前后分别34℃和37℃的变温策略研究,其平均效价合成速率为41.14 U(mL·h),效价在29 h到达最高点(1193 UmL),比对照(37℃培养)合成周期延长了8 h,效价提高了24%,也比全程34℃培养提高了9.2%。该策略增产效果显著,便于工艺控制。
Abstract:
In the process of Bacillus licheniformis fermentation of bacitracin, the strength of pre cell growth and metabolism will affect bacitracin synthesis. In order to improve the potency, the effect of variable temperature control strategy on potency synthesis was studied at the level of 20L fermentor. It was found that the potency at 37°C fermentation temperature (control) reached a high point (962 UmL) at 21h, and the average potency synthesis rate was 45.33 U(mL·h). By lowering the fermentation culture temperature (35.5℃, 34℃, 32.5℃ and 31℃), the growth rate and sugar consumption rate of the cells could be reduced. Although the average titer synthesis rate would be reduced, the titer synthesis time was extended. At the fermentation temperature of 34℃, the average titer synthesis rate of 37.66 U(mL·h) reached the high point of titer (1092 UmL) at 29h, which was an increase of 13.5% compared to 37℃. Based on the cooling control strategy, the temperature change strategies of 34℃ and 37℃ before and after 18 hours were studied. The average titer synthesis rate was 41.14 U(mL·h),and the potency reached a high point (1193 UmL) at 29h, which was 8 hours longer than the control (cultured at 37℃). The potency increased by 24%, and also increased by 9.2% compared to the whole 34℃ culture. This strategy has obvious effect of increasing production, easy process control, and can provide important reference for industrial production.
