摘要:
 动物益生菌-枯草芽胞杆菌的工业生产存在芽胞率不稳定等问题。 在30 L罐水平研究不同发酵策略对枯草芽胞杆菌生长效率的影响, 确定优化培养基与发酵条件: 玉米淀粉 35 g·L-1, 豆粕40 g·L-1, 葡萄糖5 g·L-1, 酵母浸粉3 g·L-1, 蛋白胨8 g·L-1, 轻质碳酸钙3 g·L-1, 七水硫酸镁0.5 g·L-1, 氯化钠2 g·L-1, 三水磷酸氢二钾2 g·L-1, 一水硫酸锰0.3 g·L-1, 发酵24 h温度从35℃升至40℃, 生物量(36 h)达217×108 CFU/mL, 芽胞数208亿个/mL, 较优化前分别提高28.4%、 31.6%, 为枯草芽胞杆菌工业化生产稳定芽胞率提供重要参考。
Abstract:
 The industrial production of animal probiotics Bacillus subtilis suffers from problems such as long fermentation cycle, high cost, and unstable spore rate. The effects of different fermentation strategies on the growth efficiency of Bacillus subtilis were studied at the level of 30 L tanks, and the optimal medium and fermentation conditions were determined: corn starch 35 g·L-1, soybean meal 40 g·L-1 , glucose 5 g·L-1 , yeast extract 3 g·L-1 , peptone 8 g· L-1 , light calcium carbonate 3 g·L-1 , magnesium sulfate heptahydrate 0.5 g·L -1 , sodium chloride 2 g· L -1 , dipotassium hydrogen phosphate trihydrate 2 g·L-1 , manganese sulfate monohydrate 0.3 g·L-1 , fermentation temperature increased from 35℃ to 40℃ for 24 h, biomass (36 h) reached 217×108 CFU/mL, the number of spores was 20.8 billion/mL, which were increased by 28.4% and 31.6% respectively compared with before optimization, which provided an important reference for the rate of stable spores in the industrial production of Bacillus subtilis.
