摘要:
 以新鲜活泥鳅为原料,经冷冻干燥分离得到泥鳅表皮,采用水提-醇沉法得到其活性物质。对提取温度、提取时间、浸提次数以及料液比等进行单因素试验,采用L16(45)正交试验对其提取工艺进行优化,确定最佳提取工艺。对该活性物质进行定性、定量分析,红外图谱证明本品主要构成为多糖和蛋白质;采用光度法测定其蛋白、多糖含量,结果表明泥鳅表皮中活性物质的提取率为8.52%,其中多糖含量为25%,蛋白质含量为15%。
Abstract:
 Using fresh loach as raw material, the loach epidermis was separated by freeze-drying, and its active substances were obtained by water extraction-alcohol precipitation. In this study, the extraction temperature, extraction time, extraction times and the ratio of material to liquid were tested by single factor test. On this basis, L16(45)orthogonal test was used to optimize the extraction process, and the optimal extraction process was determined, the contents of protein and polysaccharide were determined. The infrared spectrum proved that the product was mainly composed of polysaccharide and protein. And the content of protein and polysaccharide was determined by spectrophotometry. The results showed that the extraction rate of the active substance in loach epidermis was 8.52%, the content of polysaccharide and protein in the active substance were 25% and 15% respectively.
